Hydrogen/Deuterium Exchange

The Håkansson group also has an innovation track record in hydrogen/deuterium exchange (HDX) coupled with mass spectrometry.  We showed that ECD can be utilized for activation of deuterated peptides without causing hydrogen scrambling in the polypeptide melittin, thus allowing amino acid residue-specific structural information from MS/MS experiments, i.e., which amino acids change their solvent exposure as a protein structure rearranges.  Current work includes exploration of alternative MS/MS activation techniques and implementation of supercharging for improved ECD/ETD.

Figure 3. Time evolution of the isotopic distributions of the ECD product ions, c23(2+) and c22(2+), following deuterium to hydrogen exchange of melittin. The amide hydrogen exchange rate of Arginine 24 can be derived from these two consecutive ions.
